The crash happened between junctions 2 for Wolverhampton and 3 for Cosford, Shropshire.
The overturned lorry struck the central reservation barrier before coming to a halt, National Highways said.
One lane has been closed in both directions.
Drivers were brought to a slow crawl after the smash.
Traffic is now only passing the scene of the crash via the hard shoulder on the M54 eastbound, according to the agency.
Motorists are facing two-hour delays eastbound with six miles of congestion.
Lane 2 is also still closed westbound.
National Highways urged drivers to find alternative routes and plan their journeys ahead.
“A lengthy recovery and clear-up operation will be taking place,” it added.
The M54 eastbound between J3 and J2 is not expected to clear until around 12.30pm.
Normal traffic conditions are also not expected to resume until around 1pm.
Full diversion route
National Highways has urged drivers to allow extra time for their journeys and follow a diversion route after exiting the M54 eastbound at J3 for Cosford.
The alternative route is as follows:
– Exit the M54 eastbound at J3 and join the A41 northbound
– Continue on the A41 until the A5 roundabout and then take the third exit onto the A5 eastbound
– Continue on the A5 eastbound until the Galley Roundabout with the A449
– Take the third exit onto the A449 southbound
– Continue onward on the A449 southbound to then re-join the M54 eastbound at J2 near Wolverhampton
