Officers have now shut the road between J13 Bishops Tachbrook and J14 Barford – near the Oxfordshire border.
The closure will remain in place “for at least the next few hours”, warned Warwickshire Police – as drivers are stuck in long queues.
Emergency services raced to the scene just before 7.30am, according to the force.
This included an air ambulance.
An eyewitness told the Oxford Mail: “I heard the thud whilst waiting for RAC but didn’t realise what had happened until Highways ran towards it.
“They’d seen it on the way to getting to me.”
The incident wreaked havoc for those getting to work this morning.
A statement from Inrix warned commuters: “M40 Northbound blocked, queueing traffic due to accident, one vehicle involved from J14 A452 (Royal Leamington Spa) to J15 A46 Warwick By-pass (Warwick / Longbridge Island).
“Congestion to J13 Bishops Tachbrook and the Warwick Bypass.”
A police statement read: “The road is expected to be closed for at least the next few hours so please avoid the area and follow diversions where possible.”
