Police rushed to the park after receiving reports of gunshots at around 7pm.
The man, in his 40s, was treated for gunshot wounds at the scene before being rushed to hospital.
Despite the best efforts of medical teams, he was later pronounced dead.
The woman, also in her 40s, was attended to by paramedics in the park before being taken to hospital.
Detective Superintendent Oliver Richter confirmed an investigation had been launched into the shooting.
He said: “We understand this is a deeply distressing incident for those involved, the witnesses in the park and the community in the surrounding area.
“We are in the early stages of the investigation, but we believe the man and woman are known to one another.
“At this time, we are treating it as an isolated incident and there is no wider risk to the public.
“Officers are working at pace to establish the full circumstances of the incident and will continue to support those who are affected.
“A crime scene remains in place for investigation with an increased police presence.”
A forensic tent was erected in the park last night as officers continued to recover evidence.
In a statement, The Met said it was waiting for updates on the condition of the woman, who remains in hospital.
The force added: “Officers responded to reports of a shooting in Clissold Park at 19:06hrs on Wednesday, 17 September.
“A woman in her 40s was treated by paramedics from the London Ambulance Service and the London Air Ambulance.
“She was taken to hospital, where she remains and we await an update on her condition.
“A man in his 40s also received treatment for gunshot wounds.
“Despite the best efforts of paramedics, he was later pronounced deceased at hospital.
“Enquiries are ongoing to identify his next of kin.”
